Burgess reagent first prepared by E. M. Burgess in 1968, is a mild and selective dehydrating agent for secondary and tertiary alcohols and due to the amphipolar nature it is gainfully employed in a number of creative synthetic ventures. A close examination of the structure of Burgess reagent reveals that it can act as a 1,2-dipole. To the best of our knowledge, no attempts have been made to tap full synthetic potential of the amphipolar nature of this reagent and no reports on 1,3-dipolar addition to a σ-bond in acyclic systems are available in literature. In this context, we propose to unravel novel applications of Burgess reagent based on its amphipolar nature. Rich and multifaceted chemistry of nitrones form the basis of many successful chemical transformations used in attractive synthetic strategies. For the last 50 years special attention has been given to nitrones due to their successful application as building blocks in the synthesis of various natural and biologically active compounds. Our interest in nitrones stems out of its unique character: i.e. it is a 1,3-dipole exhibiting distinct nucleophilic activity. We reasoned that 1,3-dipole possessing significant nucleophilicity should react with amphipolar Burgess reagent with elimination of triethylamine to give the corresponding five-membered ring product by formal dipolar addition to a σ bond. To test this hypothesis we studied the reaction of nitrones with Burgess reagent. This thesis reveals our attempts to explore the [3+2] annulation reaction of nitrones with Burgess reagent which was found to be followed by a rearrangementinvolving C-to-N aryl migration, ultimately resulting in diarylamines and carbamates. We have also examined the reaction of cyanuric chloride with nitrones in DMF with a view to exploit the nucleophilicty of nitrones and to unravel the migratory aptitude, if any, observed in this reaction

The thesis entitled novel heterocyclic constructions mediated by nucleophilic carbenes and related chemistry, embodies the results of the investigations carried out to explore the reactivity patterns of the 1:1 zwitterions, generated in situ from various nucleophilic carbenes and DiMethyl AcetyleneDicarboxylate(DMAD) towards aldehydes and ketones. The traditional synthesis of complex organic molecules employs stepwise formation of bonds and involves multiple steps. Besides the sequential synthesis, in several instances, the desired product can also be obtained in one pot reactions of three or more starting compounds. Such reactions in which more than two starting materials react to form a product in such a way that the majority of the atoms of the starting materials can be found in the products are called multicomponent reactions(MCRs). The results of our investigations on the application of N-heterocyclic carbenes in multicomponent reaction with DMAD and aromatic aldehydes leading to the one pot synthesis of 2-oxy-maleate and furanone derivatives. It is interesting to note that dihydrofuran and lactone motifs are present in a number of biologically active natural products and other heterocyclic compounds. It is conceivable that the novel multicomponent reactions described herein will find application in the synthesis of a variety of heterocyclic compounds, and in natural product synthesis.

Plants and microorganisms provide the pharmaceutical industry with some of the most important sources of components for the research of new medications This thesis involves the study of three medicinal plants belonging to three different important families viz, Cyperus rotundus (Cyperaceae), Stereospermum colais (Bignoniaceae) as well as the well known medicinal plant Zingiber officinale (Zingiberaceae) as the third. The first chapter gives an overview of biologically active natural products with special reference to antioxidant, antidiabetic, anti-inflammatory and antimicrobial molecules from terrestrial sources. Chapter 2 of the thesis deals with the isolation of phytochemical constituents of the medicinal plant Cyperus rotundus and its antioxidant and radical scavenging potential. Chapter 3 of the thesis describes the studies on the roots of Stereospermum colais, A Bignoniaceae plant belonging to the genus Stereospermum which is used extensively. Chapter 3 of the thesis describes the studies on the roots of Stereospermum colais, a Bignoniaceae plant belonging to the genus Stereospermum which is used extensively in Ayurveda. Chapter 4 describes the biological potential of rhizomes of Zingiber officinale. Ethyl acetate extract of ginger (EAG) possessed antioxidant activity as is evident from the results of various in vitro assays compared to other extracts .In conclusion, medicinal plants Cyperus rotundus and Stereospermum colais have been analysed for their phytochemical constituents. Also, the positive results obtained from biological activity studies such as antioxidant, anti-inflammatory and antimicrobial activity on the isolated compounds/extracts add on to the medicinal properties of these plants. Apart from that, ethyl acetate extract of Zingiber officinale (ginger) rhizomes has been shown to have very good biological potential including glucose lowering and adipocyte differentiation inhibitory effect.

The development of electrochemical sensors is currently one of the active areas of research in analytical chemistry.Voltammetric sensors as an important class of electrochemical sensors are extensively used in pharmaceutical applications.In voltammetric analysis,many active compounds in dosage forms,in contrast to excipients,can be readily oxidised or reduced at the electrode surface by applying a potential.Chemically modified electrodes have great significance in the electrochemical determination of pharmaceuticals.The modification of electrode results in efficient determination of electroactive species at very lower potential without any major interferences.The present study involves the fabrication of 8 voltammetric sensors for the drugs Metronidazole Benzoate, Sulfamethoxazole, Acyclovir, Pam Chloride , Trimethoprim , Tamsulosin Hydrochloride and Ceftriaxone Sodium.Two sensors were developed for the drug tamsulosin hydrochloride while one sensor each was developed for the other drugs.

This thesis Entitled studies on the effect of toxic heavy metal mercury on the physiology and biochemistry of an estuarine crab scylla serrata (Forskal). Evaluate the toxicity of three sub lethal concentrations of mercury, viz., 0.009 mg/l, 0.02 mg/1, and 0.04 mg/l on the mud crab, Scylla serrata through bioaccumulation, and depuration studies. To characterize the biochemical responses to the sub-lethal stress of mercury in chelate muscles, abdominal muscles, hepatopancreas and gills. To study the activity pattern of acid and alkaline phosphatases in mercury-exposed crabs. Nanoparticulate drug delivery systems provide wide opportunities for solving problems associated with drug stability or disease states and create great expectations in the area of drug delivery (Bosselmann & Williams, 2012). Nanotechnology, in a simple way, explains the technology that deals with one billionth of a meter scale (Ochekpe, et al., 2009). Fewer side effects, poor bioavailability, absorption at intestine, solubility, specific delivery to site of action with good pharmacological efficiency, slow release, degradation of drug and effective therapeutic outcome, are the major challenges faced by most of the drug delivery systems. To a great extent, biopolymer coated drug delivery systems coupled with nanotechnology alleviate the major drawbacks of the common delivery methods. Chitosan, deacetylated chitin, is a copolymer of β-(1, 4) linked glucosamine (deacetylated unit) and N- acetyl glucosamine (acetylated unit) (Radhakumary et al., 2005). Chitosan is biodegradable, non-toxic and bio compatible. Owing to the removal of acetyl moieties that are present in the amine functional groups of chitin, chitosan is readily soluble in aqueous acidic solution. The solubilisation occurs through the protonation of amino groups on the C-2 position of D-glucosamine residues whereby polysaccharide is converted into polycation in acidic media. Chitosan interacts with many active compounds due to the presence of amine group in it. The presence of this active amine group in chitosan was exploited for the interaction with the active molecules in the present study. Nanoparticles of chitosan coupled drugs are utilized for drug delivery in eye, brain, liver, cancer tissues, treatment of spinal cord injury and infections (Sharma et al., 2007; Li, et a., 2009; Paolicelli et al., 2009; Cho et al., 2010). To deliver drugs directly to the intended site of action and to improve pharmacological efficiency by minimizing undesired side effects elsewhere in the body and decrease the long-term use of many drugs, polymeric drug delivery systems can be used (Thatte et al., 2005).

The scope of the work was to synthesis few biologically active derivatives of curcumin. The derivatives were prepared by altering the keto-enol centre of curcumin by various reagents. This particular reaction centre for preparing derivative was selected keeping in mind the controversy regarding the major site responsible for antioxidant mechanism of curcumin. Most of the mechanistic study done earlier was by varying the constituents in one or both of the phenol ring present in the curcumin. The alterations at the keto-enol moiety may throw an insight into the role of the diketo moiety towards the antioxidant mechanism. Since recently curcumin has been suggested as a chemotherapeutic agent for various ailments, we also decided to check the DNA intercalating property of the derivatives synthesised.

Biosurfactants are surface active compounds released by microorganisms. They are biodegradable non-toxic and eco-friendly materials. In this review we have updated the information about different microbial surfactants. The biosurfactant production depends on the fermentation conditions, environmental factors and nutrient availability. The extraction of the biosurfactants from the cell-free supernatant using the solvent extraction procedure and the qualitative and quantitative analysis has been discussed with appropriate equipment details. The application of the biosurfactant includes biomedical, cosmetic and bioremediation. The type of microbial biosurfactants include trehalose lipids, rhamnolipids, sophorolipids, glycolipids, cellobiose lipids, polyol lipids, diglycosyl diglycerides, lipoloysaccharides, arthrofactin, lichensyn A and B, surfactin, viscosin, phospholipids, sulphonyl lipids and fatty acids. Rhamnolipid biosurfactants produced by Pseudomonas aeruginosa DS10-129 showed significant applications in the bioremediation of hydrocarbons in gasoline spilled soil and petroleum oily sludge. Rhamnolipid biosurfactant enhanced the bioremediation process by releasing the weathered oil from the soil matrices and enhanced the bioavailability of hydrocarbons for microbial degradation. It is having potential applications in the remediation of hydrocarbon contaminated sites. Biosurfactants from marine microorganisms also offer great potential in bioremediation of oil contaminated oceanic environments

Given the economic importance of Jatropha curcas, and its limited availability in the wild, it would be desirable to establish plantations ofthe tree so as to obtain assured supply of raw material for extraction of phytochemicals, and seeds for production of biodiesel. However both seed propagation as well as propagation by cuttings is unsatisfactory in this tree species. Seeds have poor viability and are genetically heterozygous leading to genetic variability in terms of growth, biomass, seed yield, and oil content. Stern cuttings have poor roots and the trees are easily uprooted. Tissue culture techniques could possibly be gainfully employed in the propagation of elite plants ofJaIropha. When plant tissue is passaged through in vitro culture, there is possibility of induction of variations. An estimation of somaclonal variability is useful in a determination of culture protocols. Molecular markers could be employed to estimate the amount of variations induced in callus and regenerants by different honnonal combinations used in culture. In this context the present study aims to develop an in vitro propagation protocol for the production of plantlets and to evaluate the variation induced in callus and regenerants in comparison with mother plant by the use of molecular markers and by studying phytochemicals and bio active compounds present in callus and regenerated plants

This study focuses on the fractionation and quantification of chlorophenols, the most important and potential pollutant in this category, the distribution and seasonal dynamics of MBAS, phenols and clorophenols and development of a model to describe the chemical reactivity of the estuary are utilizing the dynamics of boron. The CES is highly influenced by various anthropogenic activities like discharge of agricultural, industrial and urban wastes operation of shipyard, oil and other transporting activities, fishing, dredging etc. Seasonal values of MBAS showed high values in the surface water during monsoon compared to premonsoon and postmonsoon. In the Cochin estuary o-chlorophenol and p-chlorophenol showed low values in the surface water compared to bottom water in the northern part of the estuary and higher values in the surface water in the southern part

Investigations on thin films that started decades back due to scientific curiosity in the properties of a two-dimensional solid, has developed into a leading research field in recent years due to the ever expanding applications of the thin films in the fann of a variety of active and passive microminiaturized components and devices, solar cells, radiation sowces and detectors, magnetic memory devices, interference filters, refection and antireflection coatings etc. [1]. The recent environment and energy resource concerns have aroused an enonnous interest in the study of materials in thin film form suitable for renewable energy sources such as photovoltaic devices. Recognition of the immense potential applications of the chalcopyrites that can fonn homojunctions or heterojunctions for solar cell fabrication has attracted many researchers to extensive and intense research on them. In this thesis, we have started with studies performed on CuInSe, thin films, a technologically well recognized compound belonging to the l•ill-VI family of semiconductors and have riveted on investigations on the preparation and characterization of compoWlds Culn3Se5. Culn5Seg and CuIn7Se12, an interesting group of compounds related to CuInSe2 called Ordered Vacancy Compounds, having promising applications in photovoltaic devices. A pioneering work attempted on preparing and characterizing the compound Culn7Sel2 is detailed in the chapters on OVC's. Investigation on valence band splitting in avc's have also been attempted for the first time and included as the last chapter in the thesis. Some of the salient features of the chalcopyrite c.ompounds are given in the next section .of this introductory chapter.
